Unser Labor beschäftigt sich mit Proteinschäden in Zellen und Geweben. Verursacher dieser Schäden sind freie Radikale und Oxidantien, die ein Nebenprodukt des Zellstoffwechsels sind. Sie entstehen verstärkt bei pathologischen Veränderungen und lösen oxidativen Stress aus. Zu diesen pathologischen Veränderungen zählen z. B. Hyperglykämie (krankhaft erhöhter Blutzucker), Hypertonie (Bluthochdruck) aber auch Inflammationen (Entzündungen).
In einer normal funktionierenden Zelle bzw. im Gewebe werden oxidativ veränderte Proteine über die intrazelluläre Proteolyse abgebaut und somit entgiftet. Diesen Mechanismus wollen wir besser verstehen. Denn in verschiedenen Stoffwechselsituationen, bei Erkrankungen oder auch im Alter funktionieren die proteolytischen Systeme oft nur unzureichend, so dass der Abbau der modifizierten Proteine unvollständig ist. Insbesondere während des Alterungsprozesses sind Zellen nicht mehr in der Lage, auf Stress adäquat zu reagieren. Es kommt zu Fehlreaktionen und zur Ansammlung dieser geschädigten Proteine in Form von Lipofuscin, einem Alterungspigment. Dieses wiederum kann die Funktion der Zelle beeinträchtigen.
Unter Zuhilfenahme modernster molekularbiologischer Methoden möchten wir herausfinden, wie Ernährung die Bildung bzw. den Abbau geschädigter Proteine beeinflusst. Dabei untersuchen wir sowohl die Effekte von Makronährstoffen, vor allem von Proteinen, als auch von Mikronährstoffen, insbesondere Vitaminen, auf den Proteinhaushalt alternder Zellen. Mit translationalen Ansätzen versuchen wir, verlässliche Biomarker zu identifizieren, die eine Beurteilung des Redox-Status und der Proteinhomöostase erlauben.

Das Verbundprojekt food4future – Nahrung der Zukunft (f4f), gefördert vom Bundesministerium für Bildung und Forschung im Rahmen des Programms Agrarsysteme der Zukunft, untersucht radikale Innovationen für eine nachhaltige und gesunde Lebensmittelversorgung.
Das Projekt wird vom Leibniz-Institut für Gemüse- und Zierpflanzenbau (IGZ) koordiniert. Das Gesamtziel von F4F ist die zukünftige, nachhaltige Sicherung ausreichender und gesunder Ernährung. Das DIfE bearbeitet das Forschungsfeld „Smart Nutrition und tragbare Sensoren“ zusammen mit dem Fachgebiet Telematik der Technischen Hochschule Wildau. In einer Humanstudie werden über einen Zeitraum von 8 Wochen neuartige Hautsensoren für die Bestimmung des Carotinoid-Status, eine eigens programmierte Carotinoid-Ernährungs-App sowie standardisierte Methoden (Blutanalysen, Fragebögen) eingesetzt und verglichen. Neuartige, in F4F entwickelte Produkte (z.B. Algen-Smoothie) sollen über Ernährungsempfehlungen durch die App in einer Folgestudie mit einbezogen werden.

Das Deutsche Zentrum für Diabetesforschung e.V. (DZD) verfolgt das Ziel, durch interdisziplinäre und translationale Forschung die Ursachen des Diabetes zu klären und neue Präventions- und Therapiekonzepte zu entwickeln.
Die Prävalenz des Typ-2-Diabetes nimmt mit dem Alter deutlich zu, der Einfluss des Alterungsprozesses bei Typ-2-Diabetes Pathogenese ist jedoch noch unzureichend verstanden. Unsere Abteilung untersucht in Kooperation mit DZD-Partnern altersassoziierte Prozesse und deren Einfluss auf die Funktion insulinproduzierender Betazellen. Mithilfe von Einzelzell-Genexpressionsanalysen (scRNA-seq) soll die Diversität alternder β-Zellen in verschiedenen Altersstufen im Zusammenhang mit der Ernährung untersucht werden. Ein weiteres Interesse besteht in der Untersuchung sogenannter Senolytika, Wirkstoffe die gezielt gealterte Zellen eliminieren können. Wir untersuchen, ob diese Wirkstoffe spezifisch gealterte β-Zellen entfernen können und die funktionellen Konsequenzen dieser „Senolyse“. Diese Daten ermöglichen einen tiefen Einblick in das Spektrum alternder β-Zellen, deren funktionelle Rolle und pathophysiologische Bedeutung für die Entwicklung eines Typ-2-Diabetes.

Das Deutsche Zentrum für Herz-Kreislauf-Forschung e.V. (DZHK) verfolgt das Ziel neue Therapien und Diagnoseverfahren zu entwickeln, um die Lebensqualität der Menschen mit Herz-Kreislauf-Erkrankungen zu verbessern und die Sterblichkeitsrate zu verringern. Die Ursachen von Herz-Kreislauf-Erkrankungen, zu denen alle Erkrankungen des Gefäßsystems und Herzens zählen, sind äußerst vielschichtig. Neben einer genetischen Veranlagung zählen besonders Bluthochdruck, hohes Cholesterin, Diabetes, Rauchen körperliche Inaktivität sowie eine ungesunde Ernährung zu den Risikofaktoren. In Deutschland sind Herz-Kreislauf-Erkrankungen die häufigste Todesursache, wobei die Inzidenz mit dem Alter ansteigt. Da Herz-Kreislauf-Erkrankungen häufig chronisch verlaufen und so über einen langen Zeitraum in die Lebensqualität der Menschen eingreifen, ist das DZHK bestrebt neue Erkenntnisse aus der Grundlagenforschung zügig und zielgerichtet in der Klinik zur Anwendung zu bringen.
Unsere Abteilung beschäftigt sich damit, den Einfluss einer hochkalorienreichen Ernährung auf die Herzfunktion und den Herzmetabolismus zu untersuchen. Mithilfe verschiedener Altersstufen und Diäten soll untersucht werden inwieweit die Ernährung die alternden Herzmuskelzellen beeinflussen und die Kontraktion beeinträchtigen kann. Ein weiteres Interesse besteht in der Analyse von Proteinaggregaten, die sich im Alter vor allem auch vermehrt in den Herzmuskelzellen anreichern, und deren direkten Effekt auf die Funktion der Zellen. Um zu untersuchen inwieweit Ernährung zu einer verbesserten Herzmuskelzellfunktion beitragen kann, erfolgen zudem Analysen zum Einfluss verschiedener Nährstoffe auf die Kontraktion der Herzmuskelzellen.

Wie Spurenelemente im Körper verstoffwechselt werden ist schon gut verstanden, jedoch weitgehend unbekannt ist, wie sich Spurenelemente unter physiologischen und pathophysiologischen Bedingungen gegenseitig beeinflussen. Da das Gleichgewicht der Spurenelemente zudem vom Geschlecht, Alter und der Nahrungsaufnahme abhängig ist, untersucht die DFG-Forschungsgruppe FOR 2558 „TraceAge“ den Status und die Interaktionen der essentiellen Spurenelemente Eisen, Mangan, Kupfer, Iod, Selen und Zink im Blut mit verschiedenen Biomarkern des Alterns. Ziel ist es alters- und geschlechtsspezifische Spurenelement-Fingerabdrücke zu erhalten, die helfen genetische sowie umweltbedingte Veränderungen zu identifizieren und eine Grundlage für eine verbesserte Vorsorge hinsichtlich gesunden Alterns und zukünftige Interventionsstudien zu schaffen.
Im Fokus unserer Abteilung steht die Untersuchung des Spurenelementes Eisen, dessen Status, Aufnahme und Metabolismus im alternden Herzen sowie der Einfluss von Eisen auf die Kontraktion der Herzmuskelzellen. Besonders in den Mitochondrien, den Energielieferanten für die Zelle, spielt Eisen eine wichtige Rolle. Auch die Analyse wichtiger Proteine des Eisenstoffwechsel, wie das Eisenspeicherprotein Ferritin oder der Transferrin-Rezeptor für die Eisenaufnahme, spielen eine wichtige Rolle, um Veränderungen im Eisenstatus zu charakterisieren. Innerhalb von „TraceAge“ untersucht die Abteilung, wie und ob sich der Eisengehalt von jungen zu den alternden Herzmuskelzellen verändert und welche Rolle proteolytische Systeme, wie das Ubiquitin-Proteasomale oder das Autophagie-Lysosomale Abbausystem bei dem Abbau der Proteine des Eisenstoffwechsels spielen. Neben der Analyse von Eisen soll zudem die Interaktion mit Kupfer und Zink, unter Betrachtung der Herzmuskelzellfunktion, Proteolyse und Markern des Alterns, untersucht werden.
